چکیده

Background :-Breast cancer is responsible for the death of millions of women worldwide every year. It is widespread in the world and Iraq that become a genuine problem for public health. The etiology of the disease is unknown. Genetic and hormonal factors are involved in the development of breast cancer and progression of the disease. Insulin like growth factor-ӏ (IGF-ӏ) have been implicated as essential in controlling cellular differentiation and tissue regeneration, through mitogenic , anti-apoptotic and increases cell migration so IGF-ӏpromotetumorigenesis. Objective:To evaluate the role of IGF-ӏ, insulin , estrogen and lipid profile in patients with breast cancer and their contribution to the pathogenesis of breast cancer. Subjects and Methods :This study included 60 patients with primary breast cancer age range (29-70) years and 60 age matched healthy controls . The subjects were selected from patients attending the Clinic Medical City/Baghdad Teaching Hospital / Oncology unit . Insulin like growth factor-I, insulin and estrogen were measured by ELISA. Results:Serum insulin like growth factor-I and insulin in breast cancer patients were significantly higher than healthy controls (p<0.01). The level of IGF-ӏ and estrogen are significantly higher in serum of premenopausal patients than the other studied groups( p<0.01). There were significantdifferences in cholesterol and low density lipoprotein (LDL) (p< 0.01), while there was no significant difference in the level of high density lipoprotein (HDL) between premenopausal patients and control (P>0.05), but there was a significant difference between postmenopausal patients and control group (P<0.01). . Conclusion :Insulin like growth factor-ӏ and estrogen level increased in the breast cancer in premenopausal patients , these results suggested that levels of serum IGF-I and estrogen together can be used as a predictive markers for breast cancer in the premenopausal women. Key word:Insulin like growth factor ӏ , Insulin, Estrogen, Breast cancer

ANTISENSE RNA TO THE TYPE I INSULIN-LIKE GROWTH FACTOR RECEPTOR REVERSED THE TRANSFORMED PHENOTYPE OF PC-3 HUMAN PROSTATE CANCER CELL LINE IN VITRO

The insulin-like growth factor I receptor (IGF-IR) plays an essential role in the establishment and maintenance of transformed phenotype. Interference with the IGF-IR pathway by antisense causes reversal of the transformed phenotype in many rodent and human tumor cell lines. We stably transfected the PC-3 human prostate cancer cell line with an IGF-IR antisense RNA expression plasmid. The ...

Leptin in Breast Cancer: Its Relationship with Insulin, Estrogens and Oxidative Stress

Breast cancer is the most common cancer in women. Several risk factors such as age, family history of breast cancer, marital status, early menarche and late menopause are related to breast cancer. Obesity is also a main health problem associated with breast cancer incidence and subsequent mortality. Association between obesity and expansion of breast cancer may be due to excessive sex steroid h...

Serum insulin-like growth factors, insulin-like growth factor binding proteins, and breast cancer risk in postmenopausal women.

BACKGROUND Studies have shown a positive association between serum insulin-like growth factor (IGF)-I and breast cancer risk in premenopausal but not postmenopausal women. IGF-II and estrogen receptor (ER) status has never been investigated. We examined the association between IGF-I, IGF-II, IGF binding protein (IGFBP)-2, IGFBP-3, and IGFBP-3 protease activity and breast cancer risk in postmeno...

Effect of raloxifene on insulin-like growth factor-I, insulin-like growth factor binding protein-3, and leptin in premenopausal women at high risk for developing breast cancer.

Elevated insulin-like growth factor I (IGF-I) is associated with an increased risk for developing breast cancer in premenopausal women, whereas lower leptin levels have been documented in premenopausal breast cancer cases. We determined the effect of raloxifene on IGF-I, insulin-like growth factor binding protein 3 (IGFBP-3), and leptin in premenopausal women at high risk for developing invasiv...

Relationships between plasma insulin-like growth factor-I and insulin-like growth factor binding protein-3 and second breast cancer risk in a prevention trial of fenretinide.

PURPOSE High circulating insulin-like growth factor (IGF) -I and/or low IGF-binding protein (IGFBP) -3 levels are associated with increased breast cancer risk in unaffected premenopausal women. We determined whether IGF-I and IGFBP-3 predict second breast cancer risk, and whether their changes during fenretinide explain observed reductions in second breast cancer in women </=50 years of age. ...

Insulin-like growth factor-I, soy protein intake, and breast cancer risk.

Previous studies have found that estrogen enhances the effect of insulin-like growth factor-I (IGF-I) levels on breast cancer cell growth. Participants in the Shanghai Breast Cancer Study (SBCS) consumed large amounts of soy that was high in isoflavones, which act as weak estrogens and as anti-estrogens. We assessed whether soy protein intake modified the effect of IGF-I levels on breast cancer...
